Although the analysis of the GNAQ p.Gly48Leu mutation predicted the activation of the protein function and consequently of the MEK pathway, strongly supporting the use of trametinib, we cannot exclude another mechanism by which trametinib might have contributed to stabilize the patient’s situation, including for instance a possible impact on the tumor microenvironment in melanoma, as mentioned by Kuske et al. [30]. This evidence concerns the gene MAP2K7 and melanoma.
