Simtuzumab, a monoclonal antibody against LOXL‐2, failed to demonstrate benefit in patients with either bridging fibrosis or compensated cirrhosis.98 Selonsertib, an ASK1 inhibitor, was not superior to placebo in improving fibrosis in patients with both bridging fibrosis and cirrhosis.99 Emricasan, a pan‐caspase inhibitor, did not meet the primary endpoints of improvement in portal hypertension or fibrosis in pre‐cirrhotic NASH patients.100 Despite the failures, some of these drugs are being tested in subgroups of NASH patients, or in combination regimens with other agents (see below). This evidence concerns the gene MAP3K5 and metabolic dysfunction-associated steatohepatitis.
