SLC12A1 and Hypokalemia: Loop diuretics can induce hypokalaemia by blocking the Na+-K+-2Cl- co-transporter (NKCC2) in the thick ascending limb of the loop of Henle which results in failure of sodium, potassium and chloride to be reabsorbed into the concentrated medullary interstitium (this transporter normally reabsorbs about 25% of the sodium load).
