NEK7 was required for caspase-1 processing and IL-1β release in murine macrophages containing either wild-type or hyperactive CAPS-associated mutant NLRP3-R258W (human NLRP3-R260W), indicating NEK7 regulates the inflammasome at or just downstream of NLRP3. The gene discussed is NLRP3; the disease is cryopyrin-associated periodic syndrome.
