Of all candidate mechanisms, one is currently in clinical trials, namely the inhibitor of mTOR (mammalian target of rapamycin) signaling Everolimus (in progress phase 2 trial for cortical hyperexcitability in tuberous sclerosis complex (TSC) and focal cortical dysplasia (FCD) (ClinicalTrials.gov Identifier: NCT02451696); a completed phase 3 trial with patients with TSC who have refractory partial-onset seizures (ClinicalTrials.gov Identifier: NCT01713946). Here, MTOR is linked to tuberous sclerosis.
