We examined whether downstream target genes of Wnt/β-catenin signaling are inhibited by cinobufagin in melanoma cells and found that 100 nM of cinobufagin significantly decreased c-Myc, Cyclin D1, and Axin-2 mRNA levels in LEF1-high expressing melanoma (A375 and A2058) but not in LEF1-low expressing lung cancer (A549) cells (Figure 4A). The gene discussed is MYC; the disease is lung cancer.
