Table 1 lists selected genes showing their genomic localization and reported functions. Interestingly, in addition to STAT3 itself (located at 17q21), several genes are regulatorily connected with STAT3, variously reported as activators (AURKA, BCL3, BRD2, JAK3, KPNB1, NFAT5, PIM3, ROCK1, SIX1, TPX2), inhibitors (WWOX), and targets (BATF3, HLX, IRF4, miR135b, NFATC1, RORC, VMP1/miR21). Accordingly, siRNA-mediated knockdown of STAT3 in ALCL cell line SU-DHL-1 confirmed that IRF4 and miR135b represent STAT3 target genes [40, 43] while HHEX escaped regulation by STAT3 (Supplementary Figure 8). The gene discussed is NFAT5; the disease is anaplastic large cell lymphoma.
