MLKL and cancer: Since the expression of RIP3 is lost/reduced in a large number of cancer cells/tumor tissues due to DNA methylation of the RIPK3 genomic sequence22,31,32, it is fortunate that this is the case, since a therapeutic reduction of MLKL in cancer cells may still mediate increased cancer cell death in these cancers, making MLKL inhibition a potential therapeutic strategy for cancer treatment in the presence of TRAIL pathway activators.
