When evaluating betweenness and closeness centrality by the forward genetics strategy (GO network), six genes were selected: AKT1, RAF1, LIMK1, BRAF, EGFR, and PTEN. In the other analysis, the reverse genetics approach (evaluating the HPO network) provided six genes as well: PAK1, VCP, AKT1, SMARCA4, RAF1, and PTEN. Together, the strategies provided nine candidate genes for neurofibromatosis phenotype modifiers. Here, PAK1 is linked to neurofibromatosis.
