As demonstrated by western blot analysis, deoxyshikonin treatment restricted the phosphorylation of Akt, GSK-3β, mTOR, p70S6K, and 4EBP1 in a concentration-dependent manner but caused no noticeable change on the total protein levels of Akt, GSK-3β, mTOR, p70S6K, and 4EBP1 in THP-1 and HL60 cells (Figures 6A–C), indicating that deoxyshikonin inactivated the Akt/mTOR pathway in AML cells. This evidence concerns the gene RPS6KB1 and acute myeloid leukemia.
