When we cloned the 3′UTR fragment containing the putative miR-578-binding sites downstream of a luciferase coding sequence, the cotransfection of the luciferase reporter (HIF1A-3′UTR-WT) and miR-578 mimic into the two BC cell lines produced lower luciferase activity than in cell lines cotransfected with the miR-NC control (Fig. 7b, c).
