MEFV mutations differ in correlation with disease severity; E148Q is recognized to have a milder clinical course,35 while M694V is associated with a more severe disease form and a higher risk of amyloidosis than other mutations.36 It is important to note that some authors considered E148Q as a functional polymorphism and not a disease-causing mutation.37,38 However, the role of E148Q mutation in the pathogenesis of FMF remains still unclear. The gene discussed is MEFV; the disease is familial Mediterranean fever.
