Vamorolone, a partial agonist of the glucocorticoid receptor, has been shown to be a potent NF‐κB inhibitor in preclinical mdx and in vitro studies [38], has shown dose‐responsive normalization of serum pro‐inflammatory proteins in DMD boys [39], and has shown preliminary evidence of improvement of muscle strength and endurance in an open‐label clinical trial [40] (Fig. 1B). This evidence concerns the gene NFKB1 and Duchenne muscular dystrophy.
