TRIM45 directly interacts with the receptor for activated C-kinase (RACK1) to negatively regulate MAPK signaling by inhibiting RACK1/protein kinase C (PKC) complex formation (Sato et al. 2015) while it directly interacts with p53 by its FLMN domain to conjugate K63-linked polyubiquitination chain to stabilize and activate p53 in glioblastoma cell lines (Zhang et al. 2017).
