In this study, we have demonstrated for the first time that inhibition of both the ligand and the receptor of the HGF/c-MET pathway (using a neutralising antibody against HGF and a small-molecule c-MET receptor inhibitor) in combination with a chemotherapeutic agent (gemcitabine), that is, (triple therapy, Hi + Ci + G) significantly reduced tumour size and, more importantly, completely eradicated metastasis in a clinically relevant advanced pancreatic cancer model. The gene discussed is HGF; the disease is familial pancreatic carcinoma.
