Inhibition of G6Pase, which has been proposed to be a shared hypoglycemic mechanism of plants, presents two major problems as a therapeutic target in patients with T2D; (1) since this enzyme is present in both gluconeogenesis and glycogenolysis pathways, inhibitors may cause hypoglycemia; and (2) accumulation of intracellular G6P may induce the expression of lipogenic genes that could result in hepatic steatosis (Agius, 2007). Here, G6PC1 is linked to type 2 diabetes mellitus.
