To evaluate whether it is the ability of WNT5A to increase the expression of MARCKS or whether it is its ability to elevate the phosphorylation level of MARCKS that is crucial for melanoma cell invasion, we took a direct approach to inhibit MARCKS phosphorylation with a cell-permeable peptide identical to the MARCKS N-terminus sequence (the MANS peptide), a peptide that does not affect MARCKS expression.
