Finally, we note that our assay can also be carried out using other CoA-activated metabolites, including the branched chain amino acid-derived metabolites propionyl-CoA, isobutyryl-CoA, isovaleryl-CoA, and 2-methylbutyryl-CoA, to help gain insights into the molecular underpinning of how FASN enzyme promiscuity drives branched-chain FA synthesis in adipose tissues and contributes to type-2-diabetes and other metabolic diseases [35,36].
