To study if the novel LEO160 promoter was also able to increase the expression levels and the immune responses of a soluble antigen, such as the HIV-1 gp120 protein, in comparison with the HIV/AIDS vaccine candidate MVA-B that expresses HIV-1 gp120 under the VACV sE/L promoter [32], a novel MVA vector expressing the HIV-1 envelope gp120 protein (clade B, isolate BX08) under the control of the synthetic VACV LEO160 promoter was generated (termed MVA-LEO160-gp120), as described in Materials and Methods (Figure 1A).
