RHEB and memory impairment: Similar to the WT mice, 6-month-old 5XFAD mice injected with bilateral intrahippocampal AAV1-Rheb(S16H) at 2 months of age spent significantly more time exploring novel objects than familiar, at both 1 and 24 h (Figure 4C,D; * p < 0.05 and #p < 0.001 vs. familiar objects), suggesting that the effects of Rheb(S16H) transduction of hippocampal neurons, such as neurotrophic signaling activation, preservation of LTP, and inhibition of Aβ deposits, limited the functional loss associated with memory impairments in 5XFAD mice.
