Surprisingly, HAT1 knockdown notably reduced the assembly of histone H3/H4 onto cccDNA to form minichromosome, which was detected at 0, 2, 4, 6, 8, 10, 12 dpi (days after infection) in de novo HBV-infected dHepaRG and HepG2-NTCP cells (Figure 2A-B, Figure S3B-C), suggesting that HAT1 is critical for the assembly of histone H3/H4 onto cccDNA in the assembly of cccDNA minichromosome.
