PTK2 and cancer: FAK is known as an upstream regulator of AKT in normal tissue and models of cancer.34, 35 The mTOR pathway is activated in a subset of EWS.36, 37, 38 Previous studies have shown that inhibition of IGF‐IR leads to downregulation of AKT and mTOR phosphorylation in EWS.39 Our results showed the dual FAK and IGF‐IR inhibitor TAE226 downregulated AKT phosphorylation but not mTOR.
