Loss‐of‐function of PTCH1, an inhibitor of smoothened (SMO) oncogenes, can cause nevoid basal cell carcinoma syndrome.40 Patients with relapse caused by such a mutation were previously treated with vismodegib, a small molecular inhibitor of SMO protein.40 The frequency of p.Thr678Ile gradually increased during treatment while the tumor burden showed a short‐term drop and then a quick increase. This evidence concerns the gene PTCH1 and neoplasm.
