Although DNA methylation can significantly increase the rate of spontaneous C→T mutations at CpG dinucleotides and pathogenic variation [a nonsense mutation c.535C>T in exon 3 of FGF16 was identified by Jamsheer, et al.25(2013)] in human FGF16 results in 4-5 metacarpal fusion (OMIM 309630), we could not find that pathogenic variation in human FGF16 results in 4-5 metacarpal fusion associated with cleft palate by a comprehensive search based on PubMed, Medline, Web of Science, and Embase databases up to December 2018. Here, FGF16 is linked to cleft palate.
