Subsequently, to validate the functional consequences of this effect, we transfected MDA-MB-231 breast cancer cells with plasmids expressing GFP, GFP-tagged wild-type RAC1 (GFP–RAC1) or a fusion protein containing GFP and a RAC1 chimaera that mimics RAC1 SUMOylation (GFP–SUMO1–RAC1) and, therefore, cannot be de-SUMOylated or be affected by GA.
