Indeed, pancreatic deletion of the autophagy gene Atg5 in a model of pancreatic cancer driven by oncogenic Kras and the stochastic loss of heterozygosity of Trp53 (KrasG12D; Trp53lox/+), a condition that reproduces the stepwise human development of pancreatic cancer, increases the number of PanIN lesions, but impairs the progression of PanIN to PDAC, prolonging mice survival (42). The gene discussed is ATG5; the disease is familial pancreatic carcinoma.
