In order to confirm the expression of this model pvc-operon in Photorhabdus during an insect infection we constructed transcription-translation reporter plasmids in which the promoter regions and the first 150 bp of coding sequence of pvc1, pnf [both from PaATCC43949 PVCpnf] and the P. asymbiotica chromosomal rpsM ribosomal ‘housekeeping’ gene (as a positive control) were genetically fused in frame to a gfpmut2 gene with no start codon (referred to hereon as pvc1::gfp, pnf::gfp and rpsM::gfp reporters). This evidence concerns the gene ENSG00000280371 and infection.
