Even should some of these inhibitors show acceptable efficacy and tolerability, combination targeting will almost certainly be necessary for enduring responses: driver-targeted cancer therapies (e.g. EGFR targeting for EGFR-driven lung adenocarcinoma and BRAF targeting in BRAF-activated melanoma) yield impressive initial responses, but these responses often fail in under one or two years.
