NECAPs are known to bind other endocytic proteins containing FxDxΦ-like motifs, such as amphiphysin or Bin1 and CALM with KDs in a range of hundreds of μM as determined in NMR titrations (Ritter et al., 2007), indirectly influence CCV size via CALM (Miller et al., 2015, Ritter et al., 2013), and a loss-of-function mutation in necap1 is a heritable cause of early infantile epileptic encephalopathy (Alazami et al., 2014). This evidence concerns the gene NECAP1 and early-infantile DEE.
