In the tumor extracellular space, TSP1 and PF4 are thought to regulate the formation of pro-angiogenic signaling complexes involving VEGF and FGF2 through two different mechanisms: sequestration – binding directly to VEGF and FGF2 to prevent binding to their pro-angiogenic receptors, and competition – competing for cell-surface HSPG to inhibit the formation of pro-angiogenic complexes.
