TNF and amyotrophic lateral sclerosis: Another hypothesis suggests competition with other substrates where ADAM10 is also involved, such as the TNFα signalling pathway, which has been observed to be active in ALS (Tortarolo et al., 2017) and cause cell alterations (Olmos and Lladó, 2014); this is probably related to microglial activation, one of the first events to be observed in experimental models of the disease (Beers et al., 2011; Liao et al., 2012; Liu and Wang, 2017; Gómez-Pinedo et al., 2018).
