Intriguingly, accompanied by reduced 14–3–3 binding, BRAF p-S365 phosphorylation was significantly downregulated in melanoma cells treated with TNFα (Fig. 6c; Supplementary Fig. 9g), suggesting that K27-linked ubiquitination of BRAF may facilitate the dephosphorylation of p-S365.
