The PERK inhibitor GSK2606414 was protective in prion (Moreno et al., 2013), frontotemporal dementia (Radford et al., 2015) AD (Yang et al., 2016) and PD (Mercado et al., 2018) mouse models but secondary side effects of pancreatic toxicity appeared as well, as high levels of insulin production in the pancreas require regulation by a functional PERK pathway.
