A previous trial26 of the cholesteryl ester transfer protein inhibitor anacetrapib, which substantially increases HDL cholesterol, reported a coronary heart disease reduction that was no greater than would be expected just from the LDL-cholesterol-lowering effects of the drug, and in previous analyses27 of the China Kadoorie Biobank study, a loss-of-function variant of the cholesteryl ester transfer protein that substantially increases total HDL cholesterol had little net effect on the incidence of coronary heart disease.
