SLC12A5 and epilepsy: Considering threonine phosphorylation was shown to have opposite effects on KCC2 vs. NKCC1 activity (McCormick and Ellison, 2011), the downregulation of the WNK/SPAK/OSR1 pathway could be a very efficient mechanism to adjust neuronal Cl− homeostasis in disorders associated with altered inhibition like epilepsy, schizophrenia, autism and neuropathic pain.
