Strikingly, two of the three largest LVE loci for non-lung cancers (at or near ATXN2/BRAP and CDKN2B-AS1) show increased cancer protection associating with decreased lifespan (due to antagonistic pleiotropy with CVD), while the third (at or near MAGI3) also shows evidence of pleiotropy, having an association with CVD three times as strong as breast cancer, and in the same direction. This evidence concerns the gene CDKN2B and breast cancer.
