Because the amino acid sequence of Xenopus Rab7a is 96% identical to human Rab7a, including the four residues identified as mutated in CMT2B patients (L129F, K157N, N161T/I, and V162M), we generated four GFP-tagged pathological mutants (Rab7aK157N, Rab7aL129F, Rab7aV162M, and Rab7aN161T) and expressed them in the Xenopus RGCs. The gene discussed is RAB7A; the disease is Charcot-Marie-Tooth disease type 2B.
