While gD will preferentially bind to nectin-1 (HveC) or nectin-2 (HveB) in non-immune cells (Martinez and Spear, 2001; Richart et al., 2003), infection of immune cells is mainly thought to occur through gD-binding to HVEM (Herpes Virus Entry Mediator, TNFRSF14), a TNFR-receptor family member that interacts with numerous host ligands, both soluble and membrane-bound, and signals intracellularly depending on their specificity and orientation (cis vs. trans) (Whitbeck et al., 1997; Kovacs et al., 2009).
