To test whether caspase-4 mediated TET2-induced tumor phenotype of MCF-7 cell, we established caspase-4 knockdown MCF-7 (shCASP4-2, shCASP4-5) by short hairpin RNAs (Fig. 4a), and then performed colony formation in soft agar to determine whether caspase-4 had a role in anchorage-independent growth of MCF-7 cells.
