Mesothelin-directed CAR T cell therapy has been used widely across pre-clinical models in a variety of malignancies (51–54), including elegant studies in pancreatic cancer by Beatty et al. whereby T cells were redirected via mRNA electroporation of mesothelin-targeted CAR with CD3z and 4-1BB signaling domains to elicit a transient CAR expression for safety (55). The gene discussed is MSLN; the disease is familial pancreatic carcinoma.
