Among 26 studies providing data on seven specific cardiovascular disease outcomes, we found consistent evidence of an increased risk of venous thromboembolism in tamoxifen users compared with non-users in both randomised controlled trials and observational studies, with a correspondingly decreased risk of venous thromboembolism when aromatase inhibitor users were compared directly with tamoxifen users. The gene discussed is CYP19A1; the disease is cardiovascular disorder.
