As shown in Figure 5B, T47D cells, transfected with a specific plasmid, carrying NFκB response elements in front of the firefly luciferase gene, do not show any activation after a 24 h BAFF or APRIL incubation; contrariwise, the JNK specific inhibitor, SP600125 or shRNAs against JNK1 or JNK2, significantly blocked the effects of APRIL and BAFF on ALDH1A1 and KLF4 (Figures 5C,D), indicating that, in breast cancer cells, APRIL and BAFF, binding to BCMA, signal toward pluripotency via JNK1 and JNK2. This evidence concerns the gene MAPK9 and breast cancer.
