Classification tree analysis was then applied to these 27 biomarkers and showed that the best combination of biomarkers ensuring an increased combined sensitivity and specificity for melanoma diagnosis, is a signature of 10 AAbs, including in order, ZBTB7B, PRKCH, TP53, PCTK1, PQBP1, UBE2V1, IRF4, MAPK8_tv2, MSN and TPM1, with a sensitivity of 79%, specificity of 84% and an AUC of 0.828 (Figure 4A–4C). The gene discussed is IRF4; the disease is melanoma.
