Bortezomib, a clinically approved proteasome inhibitor has been reported to suppress NF-κB pathway in MM by various mechanisms, such as abrogating degradation and causing accumulation of IκB, thereby suppressing transcriptional activation of NF-κB (Mitsiades et al., 2002; Markovina et al., 2008). The gene discussed is NFKB1; the disease is Miyoshi myopathy.
