In order to understand if H19 high expression levels contribute to the oncogenic properties of GBM cells, we depleted H19 by siRNAs (Figure 2A and 2B and Supplementary Figure 1A) and measured cell viability and in vitro ability to migrate and invade through an artificial ECM (transwell assays).We assayed three different siRNAs targeting H19, and demonstrated that all of them efficiently knocked down H19 in both A172 and LN229 cells (Supplementary Figure 1B). The gene discussed is H19; the disease is glioblastoma.
