Serpin I1 helps cancer cells to infiltrate brain by inhibiting plasminogen activator (PA)‐plasmin system.27 In our study, sequence analysis of serpin I1 revealed a putative YAP binding site more than 6 kb upstream of the transcription start site, and it is known that YAP can bind distal enhancers through TEAD to control transcription of downstream genes.52 Using a ChIP assay, we showed that YAP binds to the serpin l1 enhancer in H2030‐BrM3 cells, and inhibiting YAP by YAP shRNA and YAP siRNA decreased serpin I1 protein and mRNA expression in H2030‐BrM3 cells. The gene discussed is SERPINI1; the disease is cancer.
