KRAS and colorectal carcinoma: This small molecule was designed to recognize and alkylate adenine residues on the template strand at codon 12 (GTT and GAT), exon 2 of mutant KRAS. The authors demonstrated that KR12 produced a selective alkylation-mediated DNA strand cleavage at the G12D/G12 V mutations and this biochemical effect is associated with a greater reduction in the proliferation of CRC cells with G12D/G12 V mutation than that observed in G12C mutant cell, since KR12 does not recognize the latter mutation.
