The in vitro addition of the HRV 3C protease to whole cell lysate resulted in the same c-terminal 23 kDa RIPK1 cleavage product (Fig. 3a), further confirming that HRV 3C is likely the viral protease responsible for cleavage of RIPK1 during HRV16 infection and this was confirmed to be the c-terminal of RIPK1 (Fig. S2).
