Combing a dual inhibitor of PI3K and mTOR with the chemotherapeutic agent doxorubicin can have very distinct effects on neuroblastoma cells depending on the sequence of application, where inhibiting PI3K signalling after inducing apoptosis leads to a higher cell death rate than blocking it prior to the addition of doxorubicin, i.e., maximal inhibition of a survival cascade does not necessarily lead to maximal therapeutic benefits66.
