While a triple combination therapy may have concerns regarding increased toxicity, emerging preclinical and clinical studies are showing the toxicity of triple combinations are tolerable, including a recent study in anaplastic thyroid cancer showed that a patient who did not respond to either mTOR (everolimus) or RAF/MEK (dabrafenib/trametinib) inhibitors alone, but did respond to the combination of all three kinase inhibitors [55–59]. This evidence concerns the gene MAP2K7 and thyroid gland undifferentiated (anaplastic) carcinoma.
